Molecular Diagnosis of HPV Infection in the Cervical Swabs from the Patients of Cervical Neoplasia

Abstract
Human papillomavirus(HPV) infection of the female genital tract has been recognized as a widespread sexually transmitted disease that is associated with venereal warts and cervical neoplasia.
The purpose of this study is to identify the prevalence of HPV infection in normal cervical tissue, cervical intraepithelial neoplasia(CIN) and invasive cervical cancer by molecular diagnosis methods, using ViraPAP HPV DNA detection kit and
polymerase
chain reaction (PCR). In this study, we analyzed HPV DNA in the cervical swabs from 80 cases of cervical tissue (32 cases of invasive cervical cancer, 10 cases of cervical intraepthelial neoplasia(CIN) and 38 cases of normal cervical tissue).
HPV DNA was more detected in 55 cases of 80 cervical swabs (68.15%) by PCR than in 33 cases of 80 cervical swabs(41.25%) by ViraPAP test (P<0.05). By ViraPAP, HPV DNA was positive in 2 of 38 normal controls (5%), 6 of 10 CINs(60%), and 25 of 32
cervical
cancers(68%). By PCR, HPV DNA was positive in 19 of 38 normal rontrals(50%), 9 of 10 CINs (90 %), and 27 of 32 cervical cancers(84%). The HPV PCR products were hybridized to diagoxigenin labelled HPV-16 probe in 21.05% of normal controls, 40% of
CINs
and 47.22% of cervical cancers. HPV-18 was positive 4 cases of cervical cancers(11.11%).
In conclusion, this study demonstrated that ViraPAP test may be an adjuvant diagnostic method differentiate the cervical pathology into non-neoplastic and neoplastic lesions. HPV PCR test is the more sensitive and accurate method than ViraPAP
test
and
has benefit to differentiate HPV subtypes. HPV-16/18 was the most predomiant virus in cervical swabs of women with normal cervical tissue and cervical neoplasia.
